How drawn water (מים שאובין) disqualifies a מקוה "according to calculation" (לפי חשבון). The גמרא first reads this as the view of רבי אליעזר בן יעקב, rejects that reading, and then רבה explains it as counting vessels, following יוסף בן חוני.

Overview

The amud explains three lines of the משנה. First, drawn water disqualifies a מקוה 'according to calculation'. The גמרא first reads this as majority plus flowing, following רבי אליעזר בן יעקב, but that reading would leave רבי יוחנן's ruling (reported by רבין) with no תנא behind it. So רבה explains that 'calculation' means counting the vessels the water came from, following יוסף בן חוני, and a ברייתא shows his view. Next, the משנה says מי חטאת are made only by putting the ashes onto the water. רבי יוחנן says this is not like רבי שמעון, who uses a גזירה שוה on the word 'עפר' to link פרה and סוטה and allows either order. The גמרא then gives the תנא of our משנה's reading of the פסוק: the item that makes the water usable always goes on top. Finally, the משנה says one בית הפרס does not create another, which goes against רבי אליעזר. The amud ends by asking how far a בית הפרס reaches according to רבנן, and the answer comes on the next amud.
